CTCF is a ubiquitously distributed chromatin binding factor that binds to DNA sequence specific sites. It contains 11 C2H2-type zinc finger regions. CTCF is involved in transcriptional regulation by binding to chromatin insulators and preventing interaction between promoter and nearby enhancers and silencers. CTCF serves as a transcriptional repressor by binding to promoters of vertebrate MYC gene and BAG1 gene. It can also serve as a transcriptional activator of APP. It also plays a critical role in gene silencing over considerable distances in the genome and preferentially interacts with unmethylated DNA, preventing spreading of CpG methylation and maintaining methylation-free zones. When bound to different DNA sequences, it can dimerize to mediate long-range chromatin looping. CTCF undergoes sumolyation on lysine 74 and 689, which contributes to its repressive function on the MYC P2 promoter.
