It is encoded by the gene mapped to human chromosome 10q24.32. It has both 17Α-hydroxylase and 17, 20-lyase activities. The protein acts as a key enzyme in the synthesis of cortisol, testosterone and oestrogen. Mutations in CYP17A1 gene are associated with mineralocorticoid excess syndromes, glucocorticoid and sex hormone deficiencies. In addition, it also increases the risk of susceptibility to prostate cancer, benign prostatic hypertrophy and occasional congenital adrenal hyperplasia (CAH). Abiraterone inhibits steroidogenic activity of CYP17A1 and retards tumor growth in castration-resistant prostate cancer (CRPC) patients.
