Description_x000D_ General description_x000D_ TRANCE (TNF-related activation-induced cytokine), also called RANKL (RANK ligand), OPGL (Osteoprotegrin ligand), and ODF (Osteoclast differentiation factor) is a member of tumor necrosis factor superfamily that is expressed on the surface of osteoblasts and is released by activated T cells. When TRANCE binds to its receptor RANK it stimulates the formation, activation, survival and adherence of osteoclasts that increases the bone resorption. These effects are inhibited when RANKL binds to OPG that leads to decrease in bone resorption process and increase in bone volume and density. The TRANCE (RANKL)/RANK/OPG collectively activates NF-κB, JNK, Akt, p38, ERK, NFATc1 pathways. This axis is vital for maintaining the bone density, dendritic cell survival, B cell differentiation, mammary gland development.
